Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004656.4(BAP1):c.1025G>T (p.Ser342Ile), citing Ambry Variant Classification Scheme 2023: The p.S342I variant (also known as c.1025G>T), located in coding exon 11 of the BAP1 gene, results from a G to T substitution at nucleotide position 1025. The serine at codon 342 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
