Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000093.5(COL5A1):c.4794C>T (p.Gly1598=), citing LabCorp Variant Classification Summary - May 2015: Variant summary: COL5A1 c.4794C>T alters a non-conserved nucleotide resulting in a synonymous change. Several computational tools predict a significant impact on normal splicing: four predict the variant creates a deep-exonic 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 6.2e-06 in 1607098 control chromosomes in the gnomAD database (v4.1 dataset). This frequency is not higher than the maximum estimated for a pathogenic variant in COL5A1 causing Ehlers-Danlos syndrome, classic type, Ehlers-Danlos syndrome, classic type, 1 (3.1e-05), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.4794C>T in individuals affected with Ehlers-Danlos syndrome, classic type, Ehlers-Danlos syndrome, classic type, 1 and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 213057). Based on the evidence outlined above, the variant was classified as uncertain significance.
