Uncertain significance for Alagille syndrome due to a JAG1 point mutation — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000214.3(JAG1):c.866G>A (p.Gly289Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 289 of the JAG1 protein (p.Gly289Asp). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with JAG1-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt JAG1 protein function.
