NM_000093.5(COL5A1):c.4934G>A (p.Cys1645Tyr) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C1645Y variant (also known as c.4934G>A), located in coding exon 62 of the COL5A1 gene, results from a G to A substitution at nucleotide position 4934. The cysteine at codon 1645 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
