Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000093.5(COL5A1):c.2398A>G (p.Ile800Val), citing Ambry Variant Classification Scheme 2023: The p.I800V variant (also known as c.2398A>G), located in coding exon 28 of the COL5A1 gene, results from an A to G substitution at nucleotide position 2398. The isoleucine at codon 800 is replaced by valine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
