NM_000071.3(CBS):c.65A>G (p.His22Arg) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H22R variant (also known as c.65A>G), located in coding exon 1 of the CBS gene, results from an A to G substitution at nucleotide position 65. The histidine at codon 22 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
