NM_006017.3(PROM1):c.303+1G>C was classified as Pathogenic by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Disruption of this splice site has been observed in individuals with autosomal recessive retinitis pigmentosa (PMID: 31129250; Invitae). This variant has been reported in individual(s) with autosomal dominant macular dystrophy (PMID: 29343940, 31129250; Invitae); however, the role of the variant in this condition is currently unclear. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. This sequence change affects a donor splice site in intron 3 of the PROM1 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in PROM1 are known to be pathogenic (PMID: 17605048, 19718270, 24154662, 25474345). This variant is not present in population databases (gnomAD no frequency).
