Pathogenic for Danon disease — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002294.3(LAMP2):c.72_96del (p.Val24_Arg25insTer), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the LAMP2 gene (transcript NM_002294.3) at coding-DNA position 72 through coding-DNA position 96, deleting 25 bases. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has not been reported in the literature in individuals affected with LAMP2-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Arg25*) in the LAMP2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in LAMP2 are known to be pathogenic (PMID: 21415759).
