This variant lies in the AXIN2 gene (transcript NM_004655.4) at coding-DNA position 1859, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 620 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with AXIN2-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Trp620*) in the AXIN2 gene. Loss-of-function variants in AXIN2 are known to be pathogenic (PMID: 21416598, 15042511). However, tissue-specific alternative splicing of AXIN2 gene results in functional isoform lacking in-frame exon 7 (also known as exon 6, PMID: 15735151).
