Pathogenic for Muscular dystrophy-dystroglycanopathy (congenital with brain and eye anomalies), type A, 7; Autosomal recessive limb-girdle muscular dystrophy type 2U — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001101426.4(CRPPA):c.1092C>A (p.Cys364Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CRPPA gene (transcript NM_001101426.4) at coding-DNA position 1092, where C is replaced by A; at the protein level this means converts the codon for cysteine at residue 364 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with ISPD-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Cys364*) in the ISPD gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ISPD are known to be pathogenic (PMID: 23288328).
