Pathogenic for Citrin deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014251.3(SLC25A13):c.70-12_72del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SLC25A13 gene (transcript NM_014251.3) at 12 bases into the intron immediately before coding-DNA position 70 through coding-DNA position 72, deleting this region. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has been observed in individual(s) with citrin deficiency (Invitae). This variant is not present in population databases (gnomAD no frequency). This variant results in the deletion of part of exon 3 (c.70-12_72del) of the SLC25A13 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in SLC25A13 are known to be pathogenic (PMID: 10369257, 14680984, 27405544).
