Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006343.3(MERTK):c.2576_2579del (p.Glu859fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts the C-terminus of the MERTK protein. Other variant(s) that disrupt this region (p.Ser860Lysfs*37, p.Asn874Ilefs*22) have been observed in individuals with MERTK-related conditions (Invitae). This suggests that this may be a clinically significant region of the protein. This premature translational stop signal has been observed in individual(s) with retinitis pigmentosa (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Glu859Valfs*36) in the MERTK gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 141 amino acid(s) of the MERTK protein.
