Uncertain Significance for Malignant hyperthermia, susceptibility to, 1 — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000540.3(RYR1):c.3235_3240dup (p.Ser1079_Tyr1080dup), citing ACMG Guidelines, 2015. This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 3235 through coding-DNA position 3240, duplicating 6 bases. Submitter rationale: This variant causes an insertion of 6 nucleotides and the in-frame insertion of 2 amino acids in the RYR1 protein. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with malignant hyperthermia susceptibility. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
