Pathogenic for X-linked Emery-Dreifuss muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000117.3(EMD):c.546T>A (p.Tyr182Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the EMD gene (transcript NM_000117.3) at coding-DNA position 546, where T is replaced by A; at the protein level this means converts the codon for tyrosine at residue 182 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the EMD protein in which other variant(s) (p.Leu217Profs*31) have been determined to be pathogenic (Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with EMD-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Tyr182*) in the EMD gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 73 amino acid(s) of the EMD protein.
