This variant lies in the RAG1 gene (transcript NM_000448.3) at coding-DNA position 1297, deleting one base; at the protein level this means shifts the reading frame starting at valine residue 433, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This variant is not present in population databases (gnomAD no frequency). For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the RAG1 protein in which other variant(s) (p.Trp959*) have been determined to be pathogenic (PMID: 11133745, 24290284, 24406074). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with RAG1-related conditions. This sequence change creates a premature translational stop signal (p.Val433Cysfs*3) in the RAG1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 611 amino acid(s) of the RAG1 protein.
