Uncertain significance for Charcot-Marie-Tooth disease dominant intermediate F — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_021629.4(GNB4):c.641G>A (p.Arg214Gln), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with GNB4-related conditions. This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 214 of the GNB4 protein (p.Arg214Gln). This variant is not present in population databases (gnomAD no frequency). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt GNB4 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
