This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 414, where C is replaced by G; at the protein level this means replaces asparagine at residue 138 with lysine — a missense variant. Submitter rationale: The p.N138K variant (also known as c.414C>G), located in coding exon 3 of the MSH2 gene, results from a C to G substitution at nucleotide position 414. The asparagine at codon 138 is replaced by lysine, an amino acid with similar properties. In a massively parallel cell-based functional assay testing susceptibility to a DNA damaging agent, 6-thioguanine (6-TG), this variant was reported to be functionally neutral (Jia X et al.
