NM_001283009.2(RTEL1):c.2690A>G (p.Lys897Arg) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K897R variant (also known as c.2690A>G), located in coding exon 28 of the RTEL1 gene, results from an A to G substitution at nucleotide position 2690. The lysine at codon 897 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
