Pathogenic for Ehlers-Danlos syndrome, dermatosparaxis type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014244.5(ADAMTS2):c.2398G>T (p.Glu800Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ADAMTS2 gene (transcript NM_014244.5) at coding-DNA position 2398, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 800 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with ADAMTS2-related conditions. This sequence change creates a premature translational stop signal (p.Glu800*) in the ADAMTS2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ADAMTS2 are known to be pathogenic (PMID: 10417273). This variant is not present in population databases (gnomAD no frequency).
