Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001077653.2(TBX20):c.88A>G (p.Lys30Glu), citing Ambry Variant Classification Scheme 2023: The p.K30E variant (also known as c.88A>G), located in coding exon 1 of the TBX20 gene, results from an A to G substitution at nucleotide position 88. The lysine at codon 30 is replaced by glutamic acid, an amino acid with similar properties. This variant has been reported in a dilated cardiomyopathy (DCM) cohort (Mazzarotto F et al. Circulation, 2020 Feb;141:387-398). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
