Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000498.3(CYP11B2):c.240-1G>T, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects an acceptor splice site in intron 1 of the CYP11B2 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in CYP11B2 are known to be pathogenic (PMID: 20494601, 22801770, 26936515). This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with aldosterone synthase deficiency (PMID: 26936515). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 2109017). Studies have shown that disruption of this splice site alters mRNA splicing and is expected to lead to the loss of protein expression (PMID: 26936515). For these reasons, this variant has been classified as Pathogenic.
