Pathogenic for Hereditary hemochromatosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003227.4(TFR2):c.2281dup (p.Ala761fs), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the TFR2 protein in which other variant(s) (p.Gly792Arg) have been determined to be pathogenic (PMID: 16424658, 26029709). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with TFR2-related conditions. This sequence change creates a premature translational stop signal (p.Ala761Glyfs*31) in the TFR2 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 41 amino acid(s) of the TFR2 protein.
