This variant lies in the ACTN2 gene (transcript NM_001103.4) at coding-DNA position 1592, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 531 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Trp531*) in the ACTN2 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in ACTN2 cause disease. This variant is present in population databases (no rsID available, gnomAD 0.007%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease.
