Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_144498.4(OSBPL2):c.313C>G (p.Gln105Glu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine, which is neutral and polar, with glutamic acid, which is acidic and polar, at codon 105 of the OSBPL2 protein (p.Gln105Glu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with OSBPL2-related conditions. This missense change has been observed in at least one individual who was not affected with OSBPL2-related conditions (Invitae). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
