Uncertain significance for Hereditary spastic paraplegia 30; Neuropathy, hereditary sensory, type 2C; Intellectual disability, autosomal dominant 9 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001244008.2(KIF1A):c.4744-16G>A, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has not been reported in the literature in individuals affected with KIF1A-related conditions. This variant is present in population databases (rs763296092, gnomAD 0.007%). This sequence change falls in intron 41 of the KIF1A gene. It does not directly change the encoded amino acid sequence of the KIF1A protein.
