Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001385125.1(OPN1SW):c.507G>C (p.Trp169Cys), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with OPN1SW-related conditions. This variant is present in population databases (rs775951590, gnomAD 0.01%). This sequence change replaces tryptophan, which is neutral and slightly polar, with cysteine, which is neutral and slightly polar, at codon 172 of the OPN1SW protein (p.Trp172Cys). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
