NM_013254.4(TBK1):c.2075A>G (p.Lys692Arg) was classified as Uncertain significance for Frontotemporal dementia and/or amyotrophic lateral sclerosis 4 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces lysine, which is basic and polar, with arginine, which is basic and polar, at codon 692 of the TBK1 protein (p.Lys692Arg). This variant is present in population databases (no rsID available, gnomAD 0.0008%). This variant has not been reported in the literature in individuals affected with TBK1-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt TBK1 protein function. Experimental studies have shown that this missense change does not substantially affect TBK1 function (PMID: 32772249). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
