Likely pathogenic for Landau-Kleffner syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001134407.3(GRIN2A):c.2326G>A (p.Asp776Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 776 of the GRIN2A protein (p.Asp776Asn). This variant is present in population databases (rs776791010, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with GRIN2A-related conditions. ClinVar contains an entry for this variant (Variation ID: 2100167). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant.
