Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032898.5(CEP19):c.11C>G (p.Thr4Ser), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This sequence change replaces threonine, which is neutral and polar, with serine, which is neutral and polar, at codon 8 of the CEP19 protein (p.Thr8Ser). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CEP19-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
