This variant lies in the KIF1A gene (transcript NM_001244008.2) at coding-DNA position 751, where G is replaced by A; at the protein level this means replaces glycine at residue 251 with arginine — a missense variant. Submitter rationale: Experimental studies have shown that this missense change affects KIF1A function (PMID: 33880452). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt KIF1A protein function. This missense change has been observed in individual(s) with autosomal dominant hereditary spastic paraplegia (PMID: 33880452). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 251 of the KIF1A protein (p.Gly251Arg). For these reasons, this variant has been classified as Pathogenic.
