This variant lies in the RDH12 gene (transcript NM_152443.3) at 43 bases into the intron immediately before coding-DNA position 344 through coding-DNA position 421, deleting this region. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the RDH12 protein in which other variant(s) (p.Ala126Val) have been determined to be pathogenic (PMID: 19140180, 30134391). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant results in the deletion of part of exon 6 (c.344-43_421del) of the RDH12 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in RDH12 are known to be pathogenic (PMID: 17964524, 22065924). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with RDH12-related conditions.
