Uncertain significance for Hereditary thrombocytopenia and hematological cancer predisposition syndrome associated with RUNX1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001754.5(RUNX1):c.453G>T (p.Met151Ile), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with RUNX1-related conditions. This variant is present in population databases (rs755340590, gnomAD 0.0009%). This sequence change replaces methionine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 151 of the RUNX1 protein (p.Met151Ile).
