Uncertain significance for Usher syndrome type 3B — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002109.6(HARS1):c.611T>C (p.Ile204Thr), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with HARS-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 204 of the HARS protein (p.Ile204Thr). This variant is not present in population databases (gnomAD no frequency).
