Pathogenic for Lymphoproliferative syndrome 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005546.4(ITK):c.380del (p.Asp127fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Asp127Valfs*138) in the ITK gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ITK are known to be pathogenic (PMID: 16860760, 22289921, 26056787). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with ITK-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic.
