Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000260.4(MYO7A):c.1357T>C (p.Cys453Arg), citing Invitae Variant Classification Sherloc (09022015): This missense change has been observed in individual(s) with clinical features of autosomal recessive deafness (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces cysteine, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 453 of the MYO7A protein (p.Cys453Arg). For these reasons, this variant has been classified as Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt MYO7A protein function.
