Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002485.5(NBN):c.321-3C>G, citing Ambry Variant Classification Scheme 2023: The c.321-3C>G intronic variant results from a C to G substitution 3 nucleotides upstream from coding exon 4 in the NBN gene. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site and may result in the creation or strengthening of a novel splice acceptor site. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
