Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.898A>G (p.Ile300Val), citing Ambry Variant Classification Scheme 2023: The p.I300V variant (also known as c.898A>G), located in coding exon 9 of the POLE gene, results from an A to G substitution at nucleotide position 898. The isoleucine at codon 300 is replaced by valine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
