Uncertain significance for Hereditary hemochromatosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003227.4(TFR2):c.2234T>G (p.Leu745Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with arginine at codon 745 of the TFR2 protein (p.Leu745Arg). The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and arginine. This variant is present in population databases (rs560223878, ExAC 0.07%). This variant has not been reported in the literature in individuals affected with TFR2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0").
