Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.6661G>A (p.Glu2221Lys), citing Ambry Variant Classification Scheme 2023: The p.E2221K variant (also known as c.6661G>A), located in coding exon 40 of the DNAH5 gene, results from a G to A substitution at nucleotide position 6661. The glutamic acid at codon 2221 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
