NM_000890.5(KCNJ5):c.834_835inv (p.Glu279Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.834_835delTGinsCA variant, located in coding exon 1 of the KCNJ5 gene, results from an in-frame deletion of TG and insertion of CA at nucleotide positions 834 to 835. This results in the substitution of the glutamic acid residue for a lysine residue at codon 279, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by BayesDel in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
