The threonine residue is highly conserved and there is a moderate physicochemical difference between threonine and isoleucine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with glutaric acidemia type I (PMID: 15248096, 17188916, 8900228). It has also been observed to segregate with disease in related individuals.
