Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 246150 control chromosomes (gnomAD). c.2675T>C has been observed in at least one individual affected with Deficiency of alpha-mannosidase (Riise_2012). The report does not provide unequivocal conclusions about association of the variant with Deficiency of alpha-mannosidase. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in 10%-<30% of normal activity (Riise_2012).
