Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001382347.1(MYO5A):c.1178A>G (p.Lys393Arg), citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt MYO5A protein function. This variant has not been reported in the literature in individuals affected with MYO5A-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces lysine, which is basic and polar, with arginine, which is basic and polar, at codon 393 of the MYO5A protein (p.Lys393Arg). This variant is not present in population databases (gnomAD no frequency).
