Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000053.4(ATP7B):c.62A>T (p.Lys21Met), citing Ambry Variant Classification Scheme 2023: The p.K21M variant (also known as c.62A>T), located in coding exon 2 of the ATP7B gene, results from an A to T substitution at nucleotide position 62. The lysine at codon 21 is replaced by methionine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
