NM_005732.4(RAD50):c.3058G>T (p.Asp1020Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1020Y variant (also known as c.3058G>T), located in coding exon 20 of the RAD50 gene, results from a G to T substitution at nucleotide position 3058. The aspartic acid at codon 1020 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
