Uncertain significance for Hepatic methionine adenosyltransferase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000429.3(MAT1A):c.475A>C (p.Lys159Gln), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This sequence change replaces lysine, which is basic and polar, with glutamine, which is neutral and polar, at codon 159 of the MAT1A protein (p.Lys159Gln). This variant is present in population databases (no rsID available, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with MAT1A-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
