This variant lies in the PURA gene (transcript NM_005859.5) at coding-DNA position 928, duplicating one base; at the protein level this means shifts the reading frame starting at threonine residue 310, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with clinical features of PURA syndrome (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Thr310Asnfs*7) in the PURA gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 13 amino acid(s) of the PURA protein.
