Uncertain significance for Congenital sideroblastic anemia-B-cell immunodeficiency-periodic fever-developmental delay syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_182916.3(TRNT1):c.856G>A (p.Asp286Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 286 of the TRNT1 protein (p.Asp286Asn). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TRNT1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
