NM_000535.7(PMS2):c.7C>G (p.Arg3Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R3G variant (also known as c.7C>G), located in coding exon 1 of the PMS2 gene, results from a C to G substitution at nucleotide position 7. The arginine at codon 3 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.

Protein context (NP_000526.2, residues 1-13): ME[Arg3Gly]AESSSTEPAK
