NM_013251.4(TAC3):c.209-1G>C was classified as Pathogenic by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Disruption of this splice site has been observed in individuals with Idiopathic hypogonadotropic hypogonadism (PMID: 20194706, 31200363). For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant is present in population databases (rs146391497, gnomAD 0.1%). This sequence change affects an acceptor splice site in intron 3 of the TAC3 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in TAC3 are known to be pathogenic (PMID: 20194706, 20332248, 25077900).
